"Couples" would be impractical if the goal is perpetuating the species.
If we were to use science to help, with a group of forty humans, the best way is to concentrate on child bearing age women and a large collection of embryos, sperm and eggs to use to impregnate those women.
Otherwise we wouldn't have the diversity necessary to avoid too much inbreeding.

An aside....
The whole notion of "race" (or sexual preference) would be out the window as genetic diversity would be a most desirable trait in succeeding generations.

A woman can't get pregnant by having sex with a Gay....He has to penetrate the vagina, which is a taboo zone for a Gay........... :dunno:
